From my experience, a colony don't go independent immediately even if it has a Quality of 2. It happens on the 2nd turn.

What if I start building a Colony Ship on 1st turn and then it goes Independent? Will I have an Independent Colony Ship then?

The answer is no, it'll kill the build and you'll lose the money invested to build the ship.
